Preparing Microscope Slides
Goal
In order to take a close look at a biological sample, you will need to prepare a microscope slide.
Materials
- 1 Microscope object slide
- 1 Microscope cover slide
- 1 Gasburner
- 1 Glass pasteur pipette or micropipette with sterile tip
- 1 Paper towel
- a droplet of demi water
- 1 inoculation loop
Method for liquid cultures
- Put the object slide on the table in front of you.
- Turn on the gasburner and heat up the glass pasteur pipette. In case you are using sterile micropipette tips, just turn on the gasburner and work close to the flame.
- Take the pipette into your primary hand and the flask with culture in the other.
- Take out the cotton plug with the pinky finger of the hand that’s holding the pipette and quickly move the opening of the flask through the flame.
- Take up a small sample with the pipette.
- Move the opening of the flask through the flame once more and put the plug back in.
- Transfer the droplet onto the object slide.
- Cover the droplet with the cover slide.
- Dispose of any excess liquid using the paper towel.
- Dispose of the used pasteur pipette.
Method for colonies on solid medium
- Put a small droplet of water on a object slide.
- Turn on the gasburner and heat up the inoculation loop until it’s red hot.
- Open up the petri dish and cool down the loop by touching the agar.
- Stick the loop into the colony.
- Swirl the loop through the droplet on the object slide.
- Cover the droplet with the cover slide.
